Analysis Tissue Expression of IFN-γ in IL-12 and/or IL-18 Gene Ablated Naïve Mice

چکیده
Interleukin 12 (IL-12) and/or interleukin 18 (IL-18) gene ablated mice were applied for the investigation of the tissue expression of interferon γ (IFN-γ). For IL-12, IL-18, IL-12/18 and wt mice, reproductive performance were recorded and IFN-γ concentrations in heart, lung, liver, spleen, kidney and serum were quantified by ELISA. There were no significant differences of IFN-γ in heart, lung and kidney between 4 strains although control group was higher. It was observed that for IL-12 mice, compared with other 3 groups, IFN-γ in liver and spleen were decreased (p < 0.05) and reproductive performance appeared to be impaired. Serum IFN-γ level of IL-12/18 mice was significantly higher (p < 0.05). It was showed that IFN-γ productions under the normal condition were independent upon IL-12 and IL-18, its expressions in various tissues were different, and optimal IFN-γ is necessary for the normal growth and development of mammals. This study is helpful for clinical cytokines therapy. Cellular & Molecular Immunology. 2005;2(1):68-72.

Effect of Feeding Oak Acorn on Expression of IL-2, IL-13 and IFN-γ Genes in Bursa Fabricius Tissue of Broiler Chickens (Short Communication)
Today, the use of oak as a replacement feed in poultry diets is well documented. However, oak contains polyphenol compounds (tannins) as antinutrient factors resulted in limited usage in the poultry rations. Generally, consumption of feeds containing tannin could affect gene expression level of immune system. Therefore, the aim of this study was to investigate the effect of different levels of ...
متن کاملComparison of Pro-inflammatory Gene Expression Profile in Spleen and Peripheral Blood Neutrophils of BALB/C Mice Infected with Leishmania Major
Introduction: Leishmaniasis is one of the common diseases between humans and animals. The present study was to investigate the expression pattern of pro-inflammatory genes in different stages of cutaneous leishmaniasis progression in order to provide a standard framework for diagnosis of various stages of the disease. Methods: In this experimental study, 2×106 Leishmania major were injected in...
متن کاملExpression of pro-inflammatory genes in lesions, spleens and blood neutrophils after burn injuries in mice treated with silver sulfodiazine
Objective(s): It is now supposed that cytokines released during the burn injuries have a great impact on the immunological and pathological responses after the burn. The main objective of this study was to investigate the expression of some pro-inflammatory genes in the wound, spleen and blood neutrophils during the healing process of burn wounds in a murine model. Materials and Methods: The ex...
متن کاملQuantiation of IL-4, IL-10 and IFN- Genes Expression after Immunization of Mice with CFP-10 and ESAT-6 Containing Vectors
Background: Tuberculosis is a disease with high morbidity, caused mainly by Mycobaterium tuberculosis (M.tb.). DNA vaccines show a promising future due to their unique advantages over conventional methods. The early-secreted antigen target (ESAT)-6 and culture filtrate protein (CFP)-10 of M.tb. antigens have been identified as vaccine candidates against Mycobacteria and used as subunit vaccines...
متن کاملEffects of 1,25-dihydroxyvitamin D3 on IL-17/IL-23 axis, IFN-γ and IL-4 expression in systemic lupus erythematosus induced mice model
Objective(s): Systemic lupus erythematosus (SLE) is a multi-factorial autoimmune disease which may be characterized by T lymphocytes dysfunctions. Th17 cells have been identified as new effector cells, which play an important role in the pathogenesis. In recent years, immunomodulatory effect of vitamin D3 has been noticed. In the present experiment, the effect of vitamin D3 on the expression of...
